Pharmacological properties
Therapeutic effect of Magnicum is the result of a synergic action of its two active ingredients that widely participate in human and animal biochemical body reactions.
Magnesium is an essential part of all enzyme reactions and ATP-dependent reactions; it participates in metabolism of carbohydrates, proteins and fats, as well as in oxidation-reduction (redox) reactions.
Magnesium activates glycolytic enzymes, oxidative enzymes, and nucleic acid synthesis enzymes; it also plays an important role myocardial reduction. Magnesium is a platelet-fibrinogen stabilizer. The pathogenetic role of magnesium deficiency in hypertension, vasospasm, neoplastic diseases and kidney stones is well recognized. Magnesium is of a great significance in neuromuscular excitation - in this process, it acts a calcium antagonist.
Pyridoxine is mostly involved in amino acid metabolism; in the form of pyridoxal phosphate it is contained in enzymes which catalyze transamination and decarboxylation , as well as contained in carbohydrates and fats. Pyridoxine facilitates amino acid transport across cell membranes; it is necessary for phosphorylase activation, formation of neurotransmitter, gamma-aminobutyric acid, glycine, and serotonin. It is involved in the exchange of vitamin B12 and folic acid, synthesis of porphyrins and unsaturated fatty acids. Pyridoxine activates metabolic processes in the myocardium, especially during hypoxia; it may have neurotropic, hematopoietic, cardiotropic and hepatotropic properties.
The mechanism of interaction between magnesium and vitamin B6 is yet unknown. Deficiency and overdose symptoms of both ingredients are the same. However the deficiency of vitamin B6 is more pronounced in simultaneous deficiency of magnesium, since magnesium is necessary for activating the PLP-dependent enzymes.
Concurrent administration of magnesium and pyridoxine increases the rate of magnesium absorption in the gut, enhances its transmembrane passage to cells due to the formation of the chelate complex of vitamin B6 - magnesium - amino acids, increases the magnesium concentration in the blood plasma and RBC, as well as reduces its excretion in the urine.
Indications
Prevention of complications caused by magnesium and/or vitamin B6 deficiency.
The drug is also used as a preventive medication in the following cases: chronic physical and mental fatigue, irritability, increased risk of stress, depression, sleep disturbances, and myalgia. In addition it is administered for preventing therosclerosis and myocardial infarction.
Magnicum is also administered in hypomagnesemia caused by continuous smoking and alcohol abuse, prolonged use of axatives, contraceptives, some diuretics or in diabetes patients with osmotic diuresis and chronic hyperglycemia.
In combined treatment of cardiovascular diseases, such as AH, congestive heart failure, cardiac arrhythmias; in patients recovering from bone fractures, and in combination therapy for osteporosis.
Use
prevention: adults and children over 12 years old – 2 tablets once a day or 1-2 tablets twice a day.
Contraindications
Hypersensitivity reactions to the drug ingredients, severe renal insufficiency (creatinine clearance <30 ml min), hypermagnesemia, vitamin B6 hypervitaminosis, AV-blockade, myasthenia gravis, Parkinson's disease (which is treated with levodopa without concomitant use of peripheral decarboxylase inhibitor), severe hypotension, and diarrhea.
Side-effects
Very rare-gastrointestinal tract disorders (abdominal pain, constipation, diarrhea, nausea, vomiting), allergic reactions (skin rash).
